目的:了解金樱子对IgA肾病肾脏功能及肾脏组织蛋白质表达的影响。方法:通过口服牛血清白蛋白(BSA)酸化水及尾静脉注射葡萄球菌肠毒素(SEB),建立IgA肾病大鼠模型,连续用药4周后,采用表面激光解吸离子化飞行时间质谱(SELDI-TOF-MS)技术扫描分析肾脏组织蛋白质的表达变化,并用生化自动仪测定血清白蛋白(Alb)、尿素氮(BUN)及肌酐(Cr)含量。结果:共扫描到70个蛋白峰,其中3个蛋白质在模型组中的表达有降低趋势,经过金樱子水醇萃取成分作用后表达增加;9个蛋白质在模型组中的表达有升高趋势,经过金樱子水醇萃取成分作用后荷质比为1033.16Da、9370.88Da的蛋白质表达降低;而分子量为7624.95Da、15229.41Da的蛋白质在治疗组中的表达高于模型组和正常组。Cr在三组中的含量分别为:正常组(24.34±2.11)μmol/l、模型组(33.61±3.01)μmol/l、治疗组(26.17±2.26)μmol/l。结论:金樱子能够调节IgA肾病大鼠肾脏组织蛋白质的表达,增加血清Cr的清除,改善肾脏功能。
Objective: To understand the effect of Rosa laevigata on renal function and renal tissue protein expression in IgA nephropathy. METHODS: A rat model of IgA nephropathy was established by oral administration of bovine serum albumin (BSA) acidified water and tail vein injection of staphylococcal enterotoxin (SEB). Four weeks after continuous administration, surface laser desorption ionization time-of-flight mass spectrometry (SELDI- The TOF-MS) technique scans for changes in protein expression in kidney tissue and uses biochemical analyzers to measure serum albumin (Alb), urea nitrogen (BUN), and creatinine (Cr) levels. RESULTS: A total of 70 protein peaks were scanned, of which 3 proteins had a decreasing trend in the model group. The expression of the three proteins increased after the effects of water-alcoholic extraction components of Rosa roxburghii; the expression of 9 proteins increased in the model group. The expression of proteins with a specific mass ratio of 1033.16 Da and 9378.88 Da decreased after the water-alcoholic components extracted from Rosa laevigata, while the proteins with molecular weights of 7624.95 Da and 15229.41 Da were higher in the treatment group than in the model group and the normal group. The contents of Cr in the three groups were: normal group (24.34±2.11) μmol/l, model group (33.61±3.01) μmol/l, and treatment group (26.17±2.26) μmol/l. Conclusion: Rosa roxburghii can regulate the expression of protein in renal tissues of rats with IgA nephropathy, increase the clearance of serum Cr, and improve renal function.
