目的:考察灯盏细辛总咖啡酸酯对大鼠局灶性脑缺血再灌注损伤的保护作用机制。方法:线栓法建立大鼠左侧大脑中动脉缺血再灌注模型;干湿重法测定脑组织含水量;取缺血侧皮层制作脑匀浆测定超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)、过氧化氢酶(catalase,CAT)活力和谷胱甘肽(glutathione,GSH)、丙二醛(malondialdehyde,MDA)含量的变化。结果:总咖啡酸酯低、中、高剂量组(20、40、60kg.mg-1)与模型组比较均能明显降低缺血再灌注大鼠脑含水量,升高SOD、GSH-PX、CAJ活力和GSH含量,同时降低MDA的含量,其中20kg.mg-1的总咖啡酸酯作用尤为显著。结论:灯盏细辛总咖啡酸酯对大鼠脑缺血再灌注损伤的保护作用,可能与其抗自由基机制有关。
Objective: To investigate the protective mechanism of Erigeron crispus total caffeate on focal cerebral ischemia-reperfusion injury in rats. Methods: The left middle cerebral artery ischemia-reperfusion model was established by thread occlusion method. The content of water in brain tissue was determined by wet-dry method. The content of superoxide dismutase (SOD) , Glutathione peroxidase (GSH-PX), catalase (CAT) and glutathione (GSH) and malondialdehyde (MDA) . Results: Compared with model group, total caffeate in low, medium and high dose groups (20,40 and 60kg.mg-1) significantly reduced brain water content, increased SOD and GSH-PX in ischemia-reperfusion rats, CAJ activity and GSH content, while reducing the MDA content, of which 20kg.mg-1 total caffeic acid ester effect is particularly significant. Conclusion: The protective effect of Erigeron cassia cassiae on cerebral ischemia-reperfusion injury in rats may be related to its anti-free radical mechanism.
