北虫草提取物影响冷应激大鼠脑组织cAMP水平与AC活性的实验研究

目的探讨北虫草提取物对冷应激大鼠不同脑组织区域环磷酸腺苷(cAMP)水平与腺苷酸环化酶(adenylate cyclase,AC)活性的影响。方法采用4℃暴露3h应激方式,持续应激4周,建立大鼠冷应激模型,用酶联免疫吸附实验(ELISA)、放射免疫分析(RIA)法检测脑皮质,脑干和丘脑下部cAMP含量和AC活性的改变。结果冷应激实验组与正常对照组比较大鼠脑皮质、脑干,丘脑下部cAMP含量和AC活性明显下降,虫草提取物能明显拮抗冷应激引起的cAMP含量和AC活性降低。结论北虫草提取物对冷应激大鼠脑组织不同区域cAMP含量和AC活性有明显的改善作用,提示持续冷应激会导致中枢神经系统功能障碍。而虫草提取物表现出的上调cAMP含量和AC活性作用可能与增强冷应激大鼠脑组织不同区域交感神经-肾上腺-靶细胞信号传导网络之间的功能有关,确切的调控机理尚需进一步深入实验研究。 Objective To investigate the effects of cordyceps militaris extract on cyclic adenosine monophosphate (cAMP) and adenylate cyclase (AC) activity in different brain regions in cold stress rats. Methods The rat model of cold stress was established by exposure to 3h stress at 4 ℃ for 4 weeks. The cortex, brainstem and hypothalamus were detected by ELISA and radioimmunoassay (RIA) Changes in cAMP content and AC activity. Results Compared with the normal control group, the cAMP content and AC activity in the cortex, brainstem and hypothalamus of rats in cold stress group were significantly decreased. Cordyceps extract significantly inhibited the cAMP content and AC activity induced by cold stress. Conclusion Cordyceps militaris extract can significantly improve cAMP content and AC activity in different regions of the brain in cold-stressed rats, suggesting that persistent cold stress may lead to central nervous system dysfunction. However, up-regulation of cAMP and AC activity induced by Cordyceps extract might be related to the function of enhancing the signal transduction network of sympathetic-adrenal-target cells in different regions of cold stress rat brain. The exact regulation mechanism needs to be further studied Experimental Study.
