目的:观察旋覆代赭汤含药血清对大鼠胃窦平滑肌细胞(SMC)IP3受体(IP3R)及钙离子(Ca2+)浓度的影响。方法:分离大鼠胃窦SMC,加入IP3受体拮抗剂肝素后,镜下观察含药血清对胃窦SMC收缩作用;检测细胞Ca2+总浓度。结果:IP3受体拮抗剂肝素可抑制旋覆代赭汤含药血清对胃窦SMC的收缩作用;各剂量组与无钙缓冲液组的细胞内钙离子总浓度相比较无明显差异。结论:旋覆代赭汤含药血清可使胃窦SMC收缩,其机制可能是通过IP3通路作用于细胞,引起细胞内质网释放Ca2+使胃窦SMCCa2+升高。
OBJECTIVE: To observe the effect of serum containing IPXR and calcium ion (Ca2 +) on the antral smooth muscle cells (SMC) of rats by Xuanfu Daizhe Tang. Methods: The SMC of gastric antrum was isolated and the heparin of IP3 receptor antagonist was added. The contraction of SMC in gastric antrum was observed microscopically. The total concentration of Ca2 + was measured. Results: IP3 antagonist heparin could inhibit the synergistic effect of the drug-containing serum of Xuankuai decoction on SMC of gastric antrum. There was no significant difference in the total intracellular calcium concentration between each dose group and the calcium-free buffer group. CONCLUSIONS: The drug-containing serum of XFXD decoction can cause the contraction of SMC in gastric antrum, which may be caused by the IP3 pathway, causing Ca2 + release from the endoplasmic reticulum and increasing SMCCa2 + in gastric antrum.
