目的研究Wnt信号通路阻断剂表没食子儿茶素没食子酸酯(epigallocatechin-3-gallate,EGCG)在颌骨牙源性角化囊性瘤开窗灌洗治疗中的调控作用。方法收集颌骨牙源性角化囊性瘤标本25例,其中开窗灌洗治疗前病例20例,开窗灌洗治疗术后病例5例,3例生理盐水灌洗,2例EGCG灌洗。应用实时荧光定量PCR及免疫组化方法,检测Wnt信号通路相关基因FZD3及MAPK10的表达,以正常口腔黏膜作为对照。结果 FZD3和MAPK10基因在颌骨牙源性角化囊性瘤中的表达高于口腔正常黏膜,应用生理盐水开窗灌洗治疗后,FZD3和MAPK10的表达降低,但仍高于正常口腔黏膜,而应用EGCG开窗灌洗治疗后,FZD3和MAPK10的表达与正常黏膜差异不大。结论 FZD3和MAPK10在颌骨牙源性角化囊性瘤中高表达,应用EGCG开窗灌洗治疗可使FZD3和MAPK10的表达降低并趋于正常。
Objective To investigate the role of Wnt signaling pathway inhibitor epigallocatechin-3-gallate (EGCG) in the treatment of mandibular odontogenic keratocystic neoplasia. Methods Twenty-five cases of mandibular odontogenic cystic neoplasms were collected, including 20 cases before windowed lavage treatment, 5 cases after open window lavage treatment, 3 cases saline lavage and 2 cases EGCG lavage . Real-time quantitative PCR and immunohistochemistry were used to detect the expression of FZD3 and MAPK10 in Wnt signaling pathway, and normal oral mucosa was used as control. Results The expression of FZD3 and MAPK10 in oral odontogenic keratocystic tumors was higher than that in oral normal mucosa. The expressions of FZD3 and MAPK10 were decreased after oral treatment with normal saline, but were still higher than those in normal oral mucosa, However, after EGCG fenestration, the expression of FZD3 and MAPK10 was not significantly different from that of normal mucosa. Conclusions FZD3 and MAPK10 are highly expressed in odontogenic keratocystic carcinomas of the jaw. The application of EGCG fenestration could decrease the expression of FZD3 and MAPK10 and tend to be normal.
