目的探讨大鼠实验性胃溃疡自愈期间胃黏膜三叶因子2(TFF2)的表达变化。方法胃前壁黏膜下注射冰乙酸,制备大鼠胃溃疡模型,免疫组织化学法及RT-PCR法分别从蛋白水平及基因水平检测正常组(6只)、溃疡组(42只)和盐水组(42只)大鼠胃黏膜组织TFF2的表达变化。结果免疫组织化学结果显示,正常组大鼠胃黏膜TFF2蛋白呈弱阳性表达。溃疡术后1d组TFF2阳性细胞即显著增多,至溃疡术后6dTFF2积分吸光度值达到高峰,阳性细胞以靠近黏膜肌层信号较强,壁细胞阳性表达较多。溃疡术后10d及14dTFF2积分吸光度值低于溃疡术后6d组,但仍维持于较高水平;溃疡术后23dTFF2积分吸光度值显著降低,但高于对照组(P<0.05或P<0.01)。RT-PCR结果显示,TFF2mRNA变化趋势与免疫组织化学结果基本一致,以溃疡术后4d和6d表达较为强烈。结论 TFF2在维持胃黏膜的完整性和促进其快速修复过程中起重要作用。
Objective To investigate the changes of gastric mucosa trefoil factor 2 (TFF2) expression in experimental gastric ulcer healing. Methods The model of gastric ulcer was established by injecting glacial acetic acid into the anterior wall of gastric antrum to establish the model of gastric ulcer in rats. Immunohistochemical and RT-PCR methods were used to detect the expression of IL-6 in normal group (6 cases), ulcer group (42 cases) and saline group (42) rat gastric mucosa TFF2 expression changes. Results Immunohistochemical results showed that TFF2 protein in gastric mucosa of normal rats was weakly positive. The TFF2-positive cells in the ulcer group increased significantly at 1 day after operation, and reached the peak value at 6 days after the ulcer operation. The positive cells showed strong signal near the muscularis mucosa and more parietal cells were positive. The absorbance value of TFF2 at 10d and 14d after ulcer was lower than that at 6d after ulcer, but still remained at a high level. The integral of TFF2 after 23d after ulcer was significantly lower but higher than that of control group (P <0.05 or P <0.01). The results of RT-PCR showed that the trend of TFF2 mRNA was basically the same as that of immunohistochemistry, and the expression of TFF2 mRNA was stronger on the 4th and 6th day after operation. Conclusion TFF2 plays an important role in maintaining the integrity of gastric mucosa and promoting its rapid repair.
