目的 :研究玄参治疗大鼠内毒素血症的血清蛋白质组变化。方法 :用双向电泳 (2DE)分析正常组、内毒素 (lipopolysaccharide ,LPS ;静脉注射 )组、玄参 (灌胃给药 )组、LPS(静脉注射 ) +玄参 (灌胃给药 )组之间的血清蛋白质组表达差异。结果 :①LPS致内毒素血症大鼠的血清在 2DE胶上 16个蛋白点 (xPr)出现非常明显的含量变化 ,与正常组比较 ,LPS组的xPr16 ,xPr19的点容量值均极显著降低 ,xPr1,xPr2 ,xPr3,xPr4 ,xPr5 ,xPr6 ,xPr7,xPr8,xPr9,xPr11,xPr12 ,xPr14 ,xPr18,xPr2 3的点容量值均极显著增高。②LPS +玄参组与LPS组比较 ,LPS引起增高的xPr1,xPr6 ,xPr7,xPr8,xPr9,xPr11,xPr12 ,xPr14 ,xPr18,xPr2 3的点容量值均显著降低。玄参在xPr1,xPr8,xPr11,xPr12 ,xPr18均表现对LPS显著的交互作用。LPS +玄参组与正常组比较 ,其中xPr8,xPr9,xPr11,xPr12 ,xPr14 ,xPr2 3蛋白点已被明显调节到正常状态 ,但是xPr18的点容量值显著低于正常组 ,xPr1,xPr6 ,xPr7的点容量值仍显著高于正常组。③玄参与LPS两因素对xPr5 ,xPr10 ,xPr19,xPr2 0 ,xPr2 1,xPr2 2均有明显的交互作用。④在玄参组 ,xPr13,xPr2 0 ,xPr2 1,xPr2 2的点容量值均比正常组显著增高 ,xPr10 ,xPr15的点容量值均比正常组显著降低 ,这 6个蛋白点在LPS组
Objective: To study the serum proteome changes of endotoxemia in rats treated with Scrophularia. Methods: Two-dimensional electrophoresis (2DE) was used to analyze the normal group, endotoxin (LPS; intravenous injection) group, Scrophulariaceae (intragastric administration) group, LPS (intravenous injection) + Scrophularia (gastric administration) group Inter serum proteome expression differences. RESULTS: 1 Serum from LPS-induced endotoxemia rats showed significant changes in the content of 16 protein spots (xPr) on 2DE gel. Compared with the normal group, the LPS group’s point capacity values of xPr16 and xPr19 were significantly reduced. The point capacitances of xPr1, xPr2, xPr3, xPr4, xPr5, xPr6, xPr7, xPr8, xPr9, xPr11, xPr12, xPr14, xPr18, xPr2 3 are significantly increased. 2 LPS + Scrophulariaceae group compared with LPS group, LPS caused by increased xPr1, xPr6, xPr7, xPr8, xPr9, xPr11, xPr12, xPr14, xPr18, xPr2 3 point capacity values are significantly reduced. Scrophulariae exhibit significant interactions with LPS at xPr1, xPr8, xPr11, xPr12, and xPr18. LPS + Scrophulariaceae group compared with the normal group, in which xPr8, xPr9, xPr11, xPr12, xPr14, xPr2 3 protein spots have been significantly adjusted to the normal state, but the xPr18 point capacity value is significantly lower than the normal group, xPr1, xPr6, xPr7 The point capacity value is still significantly higher than the normal group. The three factors involved in LPS have obvious interactions with xPr5, xPr10, xPr19, xPr2 0, xPr2 1, xPr2 2. 4 In the Scrophulariaceae group, the spot capacity values of xPr13, xPr2 0, xPr2 1, xPr2 2 were significantly higher than those in the normal group, and the xPr 10 and xPr 15 point volume values were significantly lower than those in the normal group. These 6 protein spots were in the LPS group.
