目的 探讨用EBER - 1作探针原位杂交检测淋巴瘤组织中EB病毒的意义。方法 对 16例实验性淋巴瘤组织用EBER - 1作探针原位杂交检测EB病毒 ,同时用免疫组化法检测EB病毒表达产物及电子显微镜检测EB病毒颗粒作为对照。结果 16例淋巴瘤组织中的所有肿瘤细胞EBER - 1阳性 ,定位于胞核 ,容易辨认。免疫组化检测大多数瘤细胞表达BZLF1,少数表达LMP1和EBNA2 ,电镜证实瘤细胞核内存在EB病毒颗粒。结论 用EBER - 1作探针原位检测EB病毒的方法敏感 ,特异性高 ,而且细胞定位清晰
Objective To explore the significance of detecting Epstein-Barr virus (EBV) in lymphoma tissues by in situ hybridization using EBER-1 as a probe. Methods EB virus was detected by in situ hybridization with EBER-1 as probe in 16 cases of lymphoma tissue. Epstein-Barr virus (EBV) was detected by immunohistochemistry and EB virus particles were detected by electron microscope. Results All tumor cells in 16 lymphoma tissues were positive for EBER-1, located in the nucleus and easily identified. Immunohistochemical detection of the majority of tumor cells expressed BZLF1, a small number of expression of LMP1 and EBNA2, electron microscopy confirmed the presence of EB virus particles in the tumor cell nuclei. Conclusion EBER-1 was used as a probe for in situ detection of Epstein-Barr virus. The method was sensitive, highly specific, and clearly localized.
