目的:研究大鼠口服给予马齿苋提取物后在粪便中的排泄情况。方法:采用HPLC法测定口服给药后排出的粪便生物样品,并以葛根素作为内标物,乙腈-0.1%甲酸水作为流动相梯度洗脱。本研究建立的高效液相色谱法可应用于大鼠粪便中马齿苋提取物中的咖啡酸、对香豆酸、东莨菪素、阿魏酸、槲皮素和橙皮苷的定量分析。结果:6种成分在粪便中排泄率均小于50%,可能转化成其它代谢物形式或通过其它途径排泄。结论:本实验首次建立了大鼠口服给药后以HPLC法同时测定马齿苋提取物中6种成分粪便排泄物浓度的分析方法。
OBJECTIVE: To study the excretion of exopolysaccharide in rats after oral administration of purslane extract. Methods: The fecal samples excreted after oral administration were determined by HPLC. The mobile phase consisted of puerarin as internal standard and acetonitrile - 0.1% formic acid as eluent. The established HPLC can be applied to the quantitative analysis of caffeic acid, p-coumaric acid, scopolamine, ferulic acid, quercetin and hesperidin in the excrement of Portulaca oleracea in rat feces. Results: The six components were excreted in the feces were less than 50%, may be converted to other metabolites or by other means of excretion. Conclusion: This study established the method for the simultaneous determination of fecal excretion of six components in the purslane extract by HPLC after oral administration in rats.
