目的观察卡马西平对妊娠小鼠的影响及致畸作用,并探讨叶酸对其诱发小鼠致畸效应的影响。方法小鼠随机分为3组,每组雌雄比例2∶1,分别为空白对照组(灌胃蒸馏水,10 ml/kg)、马西平组(66.7 mg/kg)、卡马西平与叶酸联合用药组(66.7 mg/kg+0.07 mg/kg)。以阴栓出现日为妊娠零天,观察小鼠有否流产、死胎、畸形、活胎数及妊娠期增重等指标。结果卡马西平可使小鼠孕期体重增量低于正常对照组(P<0.05),可使小鼠生殖能力下降且对子代产生致畸作用,叶酸对其有拮抗作用。结论对于处于妊娠期服用卡马西平治疗的癫痫患者,应给予少量叶酸可预防畸形发生。
Objective To observe the effect and the teratogenic effect of carbamazepine on pregnant mice and to explore the effect of folic acid on the teratogenic effect of the mice. Methods The mice were randomly divided into 3 groups, with the ratio of male to female being 2: 1 in each group. The mice in the control group were treated with distilled water (10 ml / kg), maclipide (66.7 mg / kg), carbamazepine and folic acid Group (66.7 mg / kg + 0.07 mg / kg). The day of vaginal suppository was zero days of pregnancy. The mice were observed for miscarriage, stillbirth, deformity, number of live births and weight gain during pregnancy. Results Carbamazepine could reduce the weight gain of pregnant mice during pregnancy compared with that of normal control group (P <0.05), and could reduce the reproductive ability of mice and teratogenic effects on offspring. Folic acid antagonized carbamazepine. Conclusion For epilepsy patients taking carbamazepine during pregnancy, a small amount of folic acid should be given to prevent deformity.
