目的 筛选受维甲酸调控的新基因片段。方法 经细胞培养 ,构建人急性早幼粒细胞白血病细胞的减数文库 ,应用半定量RT -PCR技术对减数文库的前向文库中 5 3个新基因片段进行筛选。结果 2 4个新基因片段受维甲酸调控 ,其中大多数呈上调趋势 ,少数表现为先下调后上调。结论 维甲酸诱导的基因表达效应具有明显的时相特征
Objective To screen novel gene fragments regulated by retinoic acid. Methods The subtracted cDNA library of human acute promyelocytic leukemia cells was constructed by cell culture. Fifty-three novel gene fragments in the forward library of subtractive library were screened by semi-quantitative RT-PCR. RESULTS: Twenty-four new gene fragments were regulated by retinoid acid, most of which were up-regulated. A few of them showed up-regulation first and then up-regulation. Conclusion Retinoic acid induced gene expression has obvious temporal characteristics
