目的:制备一种治疗烧烫伤的新型海藻酸钠/壳聚糖凝胶剂,并探讨该凝胶剂加入蚯蚓蛋白后对大鼠深Ⅱ度烫伤的促进愈合作用。方法:制备海藻酸钠/壳聚糖凝胶剂并检测该空白凝胶剂的基本性质,该空白凝胶剂加入提取的蚯蚓蛋白后为蚯蚓蛋白凝胶剂;采用大鼠深Ⅱ度烫伤模型,将蚯蚓蛋白凝胶剂应用于创面,观察其对大鼠深Ⅱ度烫伤创面病理变化和创面愈合的影响。测定了烫伤愈合率、愈合时间、创面含水量、胶原降解产物羟脯氨酸(OHP)含量,并对创面病理切片进行Masson三色染色。结果:制成的海藻酸钠/壳聚糖凝胶剂外观均匀、呈乳白色,兼具水化吸收双重功效,高温低温下物理化学性质稳定。蚯蚓蛋白与空白凝胶剂可均匀混合成为蚯蚓蛋白凝胶剂,创面外用蚯蚓蛋白凝胶剂可以减少早期胶原变性,降低烫伤创面水肿程度,缩短大鼠深Ⅱ度烫伤创面愈合时间。结论:海藻酸钠/壳聚糖蚯蚓蛋白凝胶剂具有促进大鼠深Ⅱ度烫伤创面愈合的作用,是一种较为理想的外用创伤制剂。
OBJECTIVE: To prepare a novel sodium alginate / chitosan gel for the treatment of scald and burn, and to explore the promotion of deep Ⅱ degree scald in rats after adding the gelatin to earthworm protein. Methods: Sodium alginate / chitosan gel was prepared and the basic properties of the blank gel were tested. The blank gel was added with earthworm protein as earthworm protein gel. The rat model of deep second degree scald The earthworm protein gel was applied to the wound to observe its effect on pathological changes and wound healing of deep partial thickness scald wounds in rats. Scald healing rate, healing time, wound moisture content, hydroxyproline (OHP) content of collagen degradation products were determined. Masson trichrome staining was performed on wound pathological sections. Results: The prepared sodium alginate / chitosan gelling agent has a uniform appearance and milky white color, which has the dual effect of hydration absorption and stable physicochemical properties at high temperature and low temperature. Earthworm protein and blank gelling agent can be uniformly mixed into earthworm protein gelatinizer. External application of earthworm protein gelatinizer can reduce early collagen denaturation, reduce the degree of scald wound edema and shorten the healing time of deep second degree scald wound in rats. Conclusion: Sodium alginate / chitosan earthworm protein gel has the effect of promoting deep second degree scald wound healing in rats and is an ideal external wound preparation.
