建立人血浆中亚胺培南浓度的高效液相色谱测定方法,探讨亚胺培南人体内药动学过程。以0.5mol/LN-2-羟乙基哌嗪-N-2-乙磺酸—50%乙二醇(体积比11)为稳定剂,以5-羟基-3-吲哚乙酸为内标,在μ-BondapakC18柱上,以0.3mol/L四乙基溴化铵乙醇液-20mmol/L磷酸氢二钾(体积比496)为流动相,检测波长298nm,对亚胺培南进行定量测定。结果血浆中亚胺培南在1~100μg/ml浓度范围内具良好线性关系,血浆回收率为98.6%±2.25%(n=6)。血浆日内及日间RSD分别为1.69%~3.76%和3.82%~6.67%。本方法简便、快捷、灵敏、准确;适用于临床血药浓度测定。
To establish a method for the determination of imipenem in human plasma by high performance liquid chromatography and to investigate the pharmacokinetics of imipenem in human. Intra-day and intra-day plasma RSD were 1.69% ~ 3.76% and 3.82% ~ 6.67%, respectively. The method is simple, rapid, sensitive and accurate; suitable for clinical serum concentration determination.
