采用酶解法提取家养梅花鹿鹿心多肽蛋白并初步检测其中的氨基酸组分。首先通过试验从木瓜蛋白酶、胰蛋白酶、胃蛋白酶和中性蛋白酶中筛选出合适的水解用酶。然后通过单因素试验和正交试验优化出最佳的液料比、酶用量和酶解时间。最后采用UPLC法初步检测鹿心酶解液中的氨基酸组分。试验结果表明:选择木瓜蛋白酶作为最佳水解酶、酶用量为2000 U/g、液料比为9:1(v/w)、酶解时间为4.5 h,在最优条件下多肽提取率为14.04%。UPLC色谱图结果显示,鹿心酶解液中含有标准样品组中的17种氨基酸,且含量比无酶解作用的鹿心提取液高很多,营养丰富,具有潜在的保健价值。
The enzymolysis method was used to extract the domestic deer deer heart proteoglycan protein and to preliminary detect the amino acid components therein. First of all, from the papain, trypsin, pepsin and neutral protease screened appropriate hydrolysis enzyme. Then, the optimal liquid to liquid ratio, enzyme dosage and enzymolysis time were optimized by single factor test and orthogonal test. Finally, UPLC method was used to detect the amino acid components in the deer heart enzymatic hydrolyzate. The results showed that papain was the best hydrolytic enzyme, the enzyme dosage was 2000 U / g, the ratio of liquid to material was 9: 1 (v / w), and the enzymolysis time was 4.5 h. Under optimal conditions, 14.04%. UPLC chromatogram results show that the deer heart enzyme solution contains 17 kinds of amino acids in the standard sample group, and its content is much higher than the deer heart extract without enzymatic action, nutritious, with potential health value.
