目的:探讨淫羊藿总黄酮对人工关节无菌性松动的防治作用机理。方法:运用MTT法检测骨水泥微粒刺激巨噬细胞(macrophage)后的自分泌物对巨噬细胞本身的影响,同时分别应用酶联免疫法及RT-PCR法检测淫羊藿总黄酮对骨水泥微粒刺激巨噬细胞分泌肿瘤坏死因子(Tumor Necrosis Factor-α,TNF-α)水平的影响。结果:骨水泥微粒刺激巨噬细胞的分泌物对巨噬细胞自身有一定的促增殖作用,并且具有一定的剂量依赖性,而RT-PCR及ELISA法则从基因转录、蛋白分泌方面证实淫羊藿总黄酮对骨水泥微粒刺激巨噬细胞具有抑制作用。结论:骨水泥微粒可刺激巨噬细胞TNF-αmRNA的表达并分泌TNF-α,而淫羊藿总黄酮能有效地抑制单核细胞TNF-αmRNA的表达,并通过下调其表达来达到抑制TNF-α的这种分泌,从而对人工关节的松动可能起到防治作用。
Objective: To investigate the preventive and therapeutic mechanism of total flavonoids of Epimedium on the aseptic loosening of artificial joints. METHODS: MTT assay was used to detect the effect of self-secretion of macrophage stimulated by cement particles on macrophage itself. Simultaneously, enzyme-linked immunosorbent assay and RT-PCR were used to detect total flavonoids of Epimedium on bone cement. Microparticles stimulate macrophage secretion of Tumor Necrosis Factor-α (TNF-α) levels. RESULTS: The secretion of macrophages stimulated by bone cement particles had certain proliferative effects on macrophages themselves and was dose-dependent, while RT-PCR and ELISA confirmed the epimedium from the aspects of gene transcription and protein secretion. Total flavonoids have inhibitory effects on macrophages stimulated by cement particles. CONCLUSION: The cement particles can stimulate the expression of TNF-α mRNA in macrophages and secrete TNF-α. The total flavonoids of Epimedium can effectively inhibit monocyte TNF-α mRNA expression and down-regulate the expression of TNF-α to inhibit TNF-α. This secretion of α may thus play a preventive role in the loosening of artificial joints.
