为研究AFT致癌的化学预防,观察了BHA和VB_2对AFT致癌作用的影响。雄性大鼠200只,分4组:AFT组,AFT+VB_2组,AFT+BHA组,对照组,各50只。AFT以二甲基亚砜(DMSO)为溶剂,腹腔注射,三个实验组注射剂量及时间相同。VB_2溶于饮水中,BHA混和于饲料中。对照组只注射DMSO。 二年后,实验组共有57只大鼠发生肿瘤,对照组无肿瘤。BHA组肝癌诱发率(13.33%)明显低于AFT组(56.25%)(P<0.05),癌前肝细胞增生性病变发生率(27.08%)及面积(0.08±0.24mm~2/cm~2)均显著低于AFT组(45.71%,2.45±7.66mm~2/cm~2)(P<0.05),肝组织γ-GT活力(2.62±0.46单位/g)也略低于AFT组(3.84±0.56)(P>0.05)。反之,VB_2组上述各项指标均略高于AFT组(P>0.05)。其他肿瘤诱发率,BHA组与VB_2组相同(41.94%)而低于AFT组(60.71%)(P>0.05)。此外,肝癌及其他肿瘤的发生时间,BHA组明显推迟。 以上表明,BHA对AFT致癌有显著抑制作用,可能成为有效的化学预防剂。
In order to study the chemoprevention of AFT carcinogenesis, the effects of BHA and VB_2 on the carcinogenicity of AFT were observed. 200 male rats were divided into 4 groups: AFT group, AFT+VB_2 group, AFT+BHA group and control group, 50 rats each. AFT was injected intraperitoneally with dimethyl sulfoxide (DMSO) as the solvent. The injection dose and time were the same in the three experimental groups. VB_2 is dissolved in drinking water and BHA is mixed in the feed. The control group was injected with DMSO only. Two years later, a total of 57 rats in the experimental group developed tumors, and the control group had no tumors. The prevalence of hepatocellular carcinoma was significantly lower in the BHA group (13.33%) than that in the AFT group (56.25%) (P<0.05). The incidence of precancerous hepatocellular hyperplasia (27.08%) and area (0.08±0.24mm~2/cm~2) ) were significantly lower than AFT group (45.71%, 2.45±7.66mm~2/cm~2) (P<0.05), and liver tissue gamma-GT activity (2.62±0.46 units/g) was also slightly lower than AFT group (3.84). ±0.56) (P>0.05). On the contrary, the above indicators in the VB_2 group were slightly higher than those in the AFT group (P>0.05). The other tumor-inducing rates were the same in the BHA group (41.94%) and the AFT group (60.71%) (P>0.05). In addition, the occurrence time of liver cancer and other tumors was significantly delayed in the BHA group. The above shows that BHA has a significant inhibitory effect on AFT carcinogenesis and may become an effective chemopreventive agent.
