目的 检测MXR7基因在人肝细胞癌组织中的表达。方法 应用32 P标记的MXR7cDNA作为探针 ,通过Northernblot分析对正常人 12种不同组织和病理证实的术前未做治疗的 7例非肝肿瘤组织及 30例肝癌和癌旁肝组织、12例正常肝组织中MXR7基因的表达进行检测。结果 MXR7mRNA在人肝脏等 12种正常组织及 7例其它部位肿瘤组织中均不表达 ;在人肝癌中呈高表达 ,而癌旁肝组织中呈低表达 ,其阳性表达率分别为 76 7%和 13 3 %。在血清AFP阴性的肝癌患者和肿瘤直径 <5cm的肝癌中 ,MXR7mRNA阳性表达率分别为 90 %和 83 3%。肝癌患者手术后 1a和 2a生存率分别是 5 9 1%和 5 4 5 %。结论 MXR7基因在人肝癌组织中的高水平表达具有普遍性、特异性和早期性 ,提示MXR7基因可以作为肝癌的生物学标志物 ,对肝穿刺组织进行MXR7基因表达检测有助于发现AFP阴性的早期肝癌。未发现MXR7mRNA阳性表达与肝癌患者的预后有明确关系
Objective To detect the expression of MXR7 gene in human hepatocellular carcinoma. Methods 32 P-labeled MXR7 cDNAs were used as probes. Northern blot analysis was performed on 12 non-hepatic tumor tissues from 12 different tissues and pathologically confirmed preoperative non-hepatic tumor tissues, 30 liver cancer tissues and adjacent liver tissues, and 12 normal individuals. The expression of MXR7 gene was detected in liver tissue. RESULTS: MXR7 mRNA was not expressed in 12 normal tissues and 7 other tumor tissues of human liver; it was highly expressed in human hepatic carcinoma and was lowly expressed in paracancerous liver tissue, with a positive expression rate of 76.7% and 13 3%. The positive rate of MXR7 mRNA expression was 90% and 83. 3% in patients with hepatocellular carcinoma with negative serum AFP and liver cancer with tumor diameter less than 5cm. The 1a and 2a survival rates of HCC patients after surgery were 5.91% and 545%, respectively. Conclusion The high-level expression of MXR7 gene in human hepatocellular carcinoma is universal, specific and early, suggesting that MXR7 gene can be used as a biomarker for liver cancer. Detection of MXR7 gene expression in liver biopsy can help to find AFP-negative Early liver cancer. No positive correlation between MXR7 mRNA expression and the prognosis of patients with liver cancer
