目的生存素是近年发现的作用最强的细胞凋亡抑制因子,通过研究姜黄素诱导胃癌细胞凋亡过程前后的生存素表达情况,探讨其用作机制。方法不同浓度的姜黄素作用于SGC-7901细胞,MTT检测细胞生长抑制率,流式细胞术检测细胞凋亡,West-ernblot分析姜黄素作用前后胃癌细胞中Survivin的表达。结果姜黄素能显著抑制体外培养的SGC-7901细胞的生长并呈量效和时效关系。流式细胞术检测到亚二倍体凋亡峰,细胞凋亡率增加。Westernblot结果提示经姜黄素作用后,Survivin表达率下降。结论姜黄素能抑制胃癌细胞SGC-7901的生长并促进其凋亡,其发生与Survivin有关。
Objective Survivin is the strongest inhibitor of apoptosis found in recent years. By studying the expression of survivin before and after curcumin induces the apoptosis of gastric cancer cells, the mechanism of action is discussed. Methods Different concentrations of curcumin were applied to SGC-7901 cells. MTT assay was used to detect the cell growth inhibition rate. Cell apoptosis was detected by flow cytometry. Survivin expression was detected by Western-ernblot in gastric cancer cells before and after curcumin administration. Results Curcumin significantly inhibited the growth of SGC-7901 cells in vitro and showed a dose-response and time-dependent manner. Flow cytometry detected sub-diploid apoptotic peak, the rate of apoptosis increased. Western blot results suggested that curcumin decreased the expression of Survivin. Conclusion Curcumin can inhibit the growth of gastric cancer cell SGC-7901 and promote its apoptosis, and its occurrence is related to Survivin.
