目的 :研究虎纹镇痛肽 1(HWAP 1)的致突变性。方法 :采用鼠伤寒沙门氏菌回复突变试验、中国仓鼠肺成纤维细胞染色体畸变试验、小鼠骨髓嗜多染红细胞微核试验研究了虎纹镇痛肽 1的致突变作用。结果 :Ames试验结果 :HWAP 1的 4个剂量组 (1μg/皿~ 10 0 0 μg/皿 ) ,在活化及非活化条件下诱发TA97、TA98、TA10 0和TA10 2菌株的回变菌落数未见明显增加 ;染色体畸变试验结果 :HWAP 1的 4个剂量组 (2 6 3μg/皿~ 2 10 0 μg/ml) ,在活化及非活化条件下CHL细胞染色体畸变率小于 5 % ;微核试验结果 :HWAP 1在 10 0 μg/皿~ 4 6 0 μg/kg剂量范围内 ,小鼠骨髓嗜多染红细胞微核率未见明显增加。结论 :在本试验条件下 ,HWAP 1无致突变作用。
Objective: To study the mutagenicity of tiger analgesic peptide 1 (HWAP 1). Methods: The mutagenic effect of tiger analgesic peptide 1 was studied by the mutation test of Salmonella typhimurium, the chromosome aberration test of Chinese hamster lung fibroblast and the micronucleus test of mouse bone marrow polychromatic erythrocytes. Results: Ames test results: 4 doses of HWAP 1 (1μg / dish ~ 100μg / dish), induced by activation and non-activation conditions TA97, TA98, TA10 0 and TA10 2 strains back to the number of strains not changed The results of chromosome aberration test showed that the chromosome aberration rate of CHL cells was less than 5% under the four dosage groups of HWAP 1 (263μg / dish ~ 2100μg / ml) under the condition of activation and non-activation. The micronucleus test RESULTS: No significant increase in micronuclei of mouse bone marrow polychromatic erythrocytes was observed in the dose range of 100 μg / ml to 460 μg / kg of HWAP 1. Conclusion: HWAP 1 has no mutagenic effect under the experimental conditions.
