目的:分析儿童川崎病患者髓过氧化物酶(myeloperoxidase,n MPO)基因单核苷酸多态性(signle nucleotide polymorphism,SNP)位点rs2333227与川崎病临床症状特点及冠状动脉扩张之间的关联。n 方法:选取73例符合诊断标准的川崎病儿童患者为研究组,81例健康体检儿童为对照组;采用限制性片段长度多态性聚合酶链反应技术检测SNP。分析n MPO基因SNP位点rs2333227基因型和等位基因频率在两组之间的差异。检测儿童川崎病冠状动脉扩张患者外周血MPO含量,分析MPO含量与冠状动脉扩张分级之间相关性。n 结果:MPO基因rs2333227位点GG基因型频率在两组之间存在差异(n χ2=4.38,n P=0.036),差异具有统计学意义。rs2333227位点G等位基因频率在研究组中高于对照组(n χ2=10.58,n P=0.001),差异具有统计学意义;rs2333227位点与颈部淋巴结肿大(n χ2=5.64,n P=0.018)、冠状动脉扩张改变(n χ2=4.60,n P=0.032)相关;MPO含量与冠状动脉扩张呈正相关性,相关系数0.705。n 结论:MPO基因rs2333227位点与儿童川崎病的发病与病情进展相关,且与儿童川崎病中的淋巴结肿大、冠状动脉扩张等症状相关,MPO含量与冠状动脉扩张具有较高正相关性。n “,”Objective:To study the association between single nucleotide polymorphism (SNP)of myeloperoxidase (n MPO) gene rs2333227and with Kawasaki diseasein childeren, and to analyze the association between n MPO and clinical symptoms and coronary artery dilatation.n Methods:73 children with Kawasaki disease were selected as the study group and 81 healthy children as the control group; SNP was detected by restriction fragment length polymorphism (RFLP) polymerase chain reaction (PCR). The genotypes and allele frequencies of rs2333227 in n MPO SNP were analyzed between the two groups.To detect the MPO content in the peripheral blood of children with Kawasaki disease and analyze the correlation between MPO content and the grade of coronary artery dilatation.n Results:The GG genotype frequency of n MPO gene rs2333227 was different between the two groups(n χ2=4.38, n P=0.036). The G allele frequency of rs2333227 in the study group was higher than that in the control group(n χ2=10.58, n P=0.001); rs2333227 was associated with cervical lymphadenopathy(n χ2=5.64, n P=0.018)and coronary artery dilatation(n χ2=4.60, n P=0.032); There was a positive correlation between MPO content and coronary artery dilatation, and the correlation coefficient was 0.705.n Conclusion:The rs2333227of n MPO gene is associated with the pathogenesis and progression of Kawasaki disease in children, and with the symptoms of lymph node enlargement and coronary artery dilatation in Kawasaki disease in children.There is a high positive correlation between MPO content and coronary artery dilatation.n
