目的 观察低分子肝素 (LMWH)对大鼠肾小球系膜细胞 (GMCs)增生及单核细胞趋化蛋白 - 1(MCP- 1)表达的作用。方法 在体外培养的大鼠GMCs株中加入脂多糖 (LPS)后 ,再分别加入不同浓度的LMWH ,设实验组 1组 :GMCs +LPS +LMWH ,设对照组 2组 :GMCs +LPS组及GMCs组 ,分别将上述 3组命名为 :LMWH组 ,LPS组及GMCs组。采用MTT掺入方法 ,于 2 4h、4 8h ,观察 3组中GMCs增生水平 ,并采用ELISA方法观察其培养的上清液中单核细胞趋化蛋白 - 1(MCP - 1)的浓度。结果 LMWH组于 4 8hGMCs增生的抑制最为显著 ,其中 :LMWH组GMCs增生水平低于LPS组 (P <0 0 5 ) ,与GMCs组无统计学差别 (P >0 0 5 ) ;LMWH组GMCs培养上清中MCP - 1浓度明显低于LPS组 (P <0 0 1) ,与GMCs组无统计学差别 (P >0 0 5 )。结论 LMWH能抑制大鼠GMCs增生 ,下调GMCs中MCP - 1的异常表达及分泌。
Objective To observe the effect of low molecular weight heparin (LMWH) on the proliferation of rat glomerular mesangial cells (GMCs) and the expression of monocyte chemoattractant protein - 1 (MCP - 1). Methods After adding lipopolysaccharide (LPS) into rat GMCs strains cultured in vitro, different concentrations of LMWH were added separately. The experimental group 1: GMCs + LPS + LMWH, control group 2: GMCs + LPS group and GMCs Group, respectively named the above three groups: LMWH group, LPS group and GMCs group. MTT method was used to observe the proliferation of GMCs in three groups at 24 h and 48 h. The concentration of monocyte chemoattractant protein - 1 (MCP - 1) in supernatants was detected by ELISA. Results The proliferation of LMWH group was the most significant at 48 h. The proliferation of GMCs in LMWH group was lower than that in LPS group (P <0.05), and there was no significant difference between the two groups (P> 0.05) The concentration of MCP - 1 in the supernatant was significantly lower than that in the LPS group (P <0.01), which was not significantly different from that in the GMCs group (P> 0.05). Conclusion LMWH can inhibit the proliferation of GMCs in rats and down - regulate the expression and secretion of MCP - 1 in GMCs.
