目的:观察HWTX-I对佐剂性关节炎大鼠抗伤害作用效果。方法:将30只W istar大鼠随机分为假手术组、非用药组和HWTX-I组。采用完全弗氏佐剂在大鼠右踝关节造成单发性关节炎炎性痛模型,假手术组注射不完全弗氏佐剂,并实施硬脊膜外腔插管手术。致炎后10d内,经由留置的硬脊膜外腔插管给实验组大鼠注射60μg/Kg体重的HWTX-I进行抗伤害作用干预,非用药组经由插管注射生理盐水,同时测定各组痛评分。10d后处死大鼠,取腰段脊髓,采用RT-PCR法测定脊髓中TNF-α表达的变化。结果:(1)HWTX-I组痛评分较非用药组呈下降趋势。(2)HWTX-I能下调大鼠腰段脊髓TNF-α表达。结论:HWTX-I可以明显改善单侧佐剂性关节炎大鼠各项痛反应指标,其可能机制是通过抑制炎性痛大鼠脊髓中相应炎性因子的表达,从而减缓痛觉过敏的形成达到减轻炎痛的目的。
Objective: To observe the anti-arthritic effect of HWTX-I in adjuvant arthritis rats. Methods: Thirty Wistar rats were randomly divided into sham operation group, non-medication group and HWTX-I group. A single arthritis inflammatory pain model was induced with complete Freund’s adjuvant in the right ankle of rats. In the sham group, incomplete Freund’s adjuvant was injected and epidural catheterization was performed. Within 10 days after inflammation, the experimental group rats were injected with 60μg / kg body weight of HWTX-I through the indwelling epidural catheter for antinociceptive effect. The non-medication group was injected with saline through cannula, and each group Pain score. After 10 days, the rats were sacrificed and the lumbar spinal cord was taken. The expression of TNF-α in the spinal cord was measured by RT-PCR. Results: (1) HWTX-I group pain scores showed a downward trend compared with non-medication group. (2) HWTX-I can down-regulate the expression of TNF-α in the lumbar spinal cord of rats. Conclusion: HWTX-I can significantly improve all kinds of pain response indicators in rats with unilateral adjuvant arthritis, and its possible mechanism is to reduce the formation of hyperalgesia by inhibiting the expression of inflammatory cytokines in the spinal cord of inflammatory pain rats Reduce the purpose of inflammation.
