目的:建立一种中药红曲基原真菌新的鉴定方法。方法:采用毛细管区带电泳分离模式,背景电解质为20 m m ol/L硼酸盐缓冲液(pH 9.11),电压20kV,温度25 ℃;紫外检测波长214 nm ,压差进样,进样时间5 s。结果:不同种红曲霉胞内和胞外水溶性蛋白质的成分和含量均存在显著差异。根据红曲霉胞内或胞外水溶性蛋白质4~10 m in 区间内的指纹图谱及这些峰的相对含量,可以有效地鉴别红曲霉属真菌,结果与形态学以及扫描电镜的检验结果一致。结论:高效毛细管电泳可用于红曲霉等真菌的分类和鉴定。
Objective: To establish a new method for the identification of red fungus-based raw fungi. Methods: The capillary zone electrophoretic separation mode was adopted. The background electrolyte was 20 m ol/L borate buffer (pH 9.11), the voltage was 20kV, the temperature was 25 °C, the UV detection wavelength was 214 nm, and the differential pressure was injected. The time is 5 s. Results: There were significant differences in the composition and content of intracellular and extracellular water-soluble proteins of different species of Monascus. According to the fingerprints of Monascus intracellular or extracellular water-soluble proteins in the range of 4-10 m in and the relative content of these peaks, Monascus fungi can be effectively identified. The results are consistent with the results of morphology and scanning electron microscopy. Conclusion: High performance capillary electrophoresis can be used for the classification and identification of fungi such as Monascus.
