目的研究基因重组人卵泡抑素(recombinant humanfollistatin,rh-FS)对实验性肝纤维化大鼠增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、溴脱氧尿苷(bromodeoxyuridine,BrDU)核标记及激活素 A(activin A,ActA)表达的影响.方法采用400 mL·L~(-1)四氯化碳(CCl_4)sc 制备大鼠实验性肝纤维化模型,将16只肝纤维化模型组大鼠随机分成 rh-FS 处理组和假手术组,另外8只大鼠不作任何处理作为正常对照.rh-FS(1μg)经大鼠回结肠静脉注入 rh-FS 处理组大鼠门静脉内,24 h 后处死动物,免疫组织化学法检测正常大鼠肝脏、rh-FS 处理组和假手术组 PCNA,BrDU 核标记及 Act A 的表达.结果 rh-FS 可明显增强肝纤维化大鼠肝细胞 PCNA(FS 处理组 PCNA 核标记19%±7%,假手术组3%±2%,n=8,P<0.01)和 BrDU(FS 处理组 DrDU 染色+,假手术组染色-,n=8)核标记,二者变化相似;肝纤维化时 Act A 表达增加(正常大鼠肝脏+,肝纤维化时++~+++),且其分布发生改变,rh-FS 短时间作用对 Act A 表达无明显影响.结论 rh-FS 可促进纤维化肝脏肝细胞增殖.
Objective To investigate the effect of recombinant human follistatin (rh-FS) on the expression of proliferating cell nuclear antigen (PCNA), bromodeoxyuridine (BrDU) A (Actin A) expression.Methods Rat experimental hepatic fibrosis model was induced by 400 mL · L -1 carbon tetrachloride (CCl 4) sc, and 16 rats with liver fibrosis The rats were randomly divided into rh-FS treatment group and sham operation group, and the other 8 rats were given no treatment as normal control.RH-FS (1μg) was injected intravenously into the portal vein of rh-FS-treated rats, The animals were sacrificed and the expression of PCNA, BrDU and Act A in normal rat liver, rh-FS treated group and sham operation group were detected by immunohistochemical method.Results rh-FS could significantly increase the expression of PCNA in liver fibrosis rats (N = 8, P <0.01), and BrDU (+ staining with DrDU in FS treatment group, staining of sham - operated group, n = 8) (P <0.05). Act A expression was increased in hepatic fibrosis (normal rat liver + and ++ ~ +++ in hepatic fibrosis) Cloth changes, rh-FS short acting Act A had no effect on expression. Conclusion rh-FS can promote hepatocyte proliferation and liver fibrosis.
