目的 探讨抗细胞凋亡因子Clusterin在膀胱癌EJ细胞中的表达及抗凋亡机制。 方法 应用免疫细胞化学染色检测Clusterin在EJ细胞中的表达 ,应用 3′末端脱氧核苷酰转移酶介导生物素标记法 (TUNEL)检测Clusterin对EJ细胞的抗凋亡作用。 结果 Clusterin在EJ细胞中有明显的阳性表达。细胞外加入Clusterin后 ,EJ细胞胞浆内染色加深 ,呈颗粒状 ,为强阳性染色。提示细胞外加入小剂量Clusterin可以明显抑制MMC诱导的EJ细胞凋亡作用。 结论 Clusterin在细胞外可以通过细胞膜进入细胞浆内发挥抗凋亡作用。检测Clusterin的表达特性有助于临床评估膀胱癌复发、耐药等生物学特性。
Objective To investigate the expression of anti-apoptotic factor Clusterin in bladder cancer EJ cells and its anti-apoptotic mechanism. Methods The expression of Clusterin in EJ cells was detected by immunocytochemical staining. The anti-apoptotic effect of Clusterin on EJ cells was detected by 3’-terminal deoxynucleotidyl transferase-mediated biotin labeling (TUNEL). Results Clusterin had obvious positive expression in EJ cells. After extracellular addition of Clusterin, EJ cells cytoplasm stained darker, granular, strongly positive staining. It suggested that low dose of Clusterin could significantly inhibit MMC induced EJ cell apoptosis. Conclusion Clusterin can play an anti-apoptotic role in the extracellular cytoplasm through the cell membrane. Detection of Clusterin expression characteristics help clinical evaluation of bladder cancer recurrence, resistance and other biological characteristics.
