目的探讨葛根及其配伍对动能性便秘的治疗机制。方法将75只SPF等级8周龄ICR雄性小鼠分为5组,包括空白组、模型组、葛黄组、葛柴组、大黄组,每组15只。空白组不做任何处理,常规灌胃生理盐水;针对模型组、葛黄组、葛柴组、大黄组,将小鼠自身粪便少许磨碎,加生理盐水制成10%的混悬液灌胃3天,从第4天开始,各组分别灌胃5天后处死,免疫组化分法观察Cajal间质细胞的表达。结果与模型组比较,葛黄组、葛柴组、大黄组小鼠胃肠细胞Cajal间质细胞黏膜表面、肌层表面等改善明显。结论以葛根为主的配伍组合可能通过对胃肠细胞Cajal间质细胞的干预来治疗功能性便秘。
Objective To explore the therapeutic mechanism of Pueraria and its compatibility on kinetic constipation. Methods 75 male ICR mice of 8 weeks old with SPF grade were divided into 5 groups, including blank group, model group, Gehuang group, GECHI group and rhubarb group, 15 in each group. For the model group, Gehuang group, Ge Chai group and Rhubarb group, mice were given a small amount of excrement, and 10% suspension of normal saline was given to the rats in the blank group. On day 3, starting from day 4, the rats in each group were sacrificed 5 days after gavage, and the expression of Cajal interstitial cells was observed by immunohistochemistry. Results Compared with the model group, the mucosal surface and muscular surface of Cajal interstitial cells of GeGroup, GeChai group and Rhubarb group mice were significantly improved. Conclusion Combination of Pueraria-based compatibility may treat functional constipation through the intervention of gastrointestinal Cajal stromal cells.
