利用农杆菌LBA4404(pTOK233)转化水稻成熟胚愈伤组织,通过筛选获得抗性愈伤组织并再生出转化植株,转化频率为8.9%。转基因植株经分子生物学鉴定为阳性,外源基因gus在转化植株中表达亦提供了转化成功的证据。转基因植株的当代及T1代植株获得了种子。摸索了应用农杆菌转化水稻的方法、条件。利用干燥处理法抑菌并提高转化植株再生频率。
Agrobacterium tumefaciens LBA4404 (pTOK233) was used to transform the callus of rice mature embryos. Resistant calli were screened and transformed into plants. The transformation frequency was 8.9%. Transgenic plants were identified as positive by molecular biology and the expression of the exogenous gene gus in transformed plants provided evidence of successful transformation. Contemporary and T1 generation plants of transgenic plants obtained seeds. The methods and conditions of using Agrobacterium to transform rice were explored. Bacteriostasis by drying treatment and increase the regeneration frequency of transformed plants.
