目的探讨建立小鼠非酒精性脂肪肝动物模型的最佳饲料配比方案,为进一步开展脂肪肝及其治疗的相关研究提供有价值的实验数据。方法 80只KM小鼠随机分为4组,甲、乙、丙为模型组,丁组为对照组。模型组给予不同梯度的高脂饮食,对照组给予正常饮食。在造模后每1周分别取模型组动物各5只处死,观察其病理模型形成情况,做血液指标检测及病理切片;采用全自动生化分析仪检测生化指标;取肝脏,制肝匀浆,测定LTG、LTC。结果造模过程中,模型组动物乙组造模后体重增长较正常对照组及模型组甲组、丙组迅速。至实验结束时,甲组体重较正常组平均重7.62%,乙组体重较正常组平均重16.76%,丙组体重较正常组平均重11.14%,模型动物体重及肝指数均较正常组显著增加(P<0.01)。生化指标检测显示模型组小鼠的TC、TG、ALT、AST、LDL-C、LTG、LTC水平,均有一定程度的降低(P<0.01),HDL-C水平显著升高(P<0.01),其中乙组小鼠的变化幅度最大(P<0.01)。在病理方面,造模组1周后乙组小鼠最早出现了轻度的肝脂肪变,造模2周乙组小鼠多呈中度脂肪变。造模4周乙组小鼠呈中重度的大泡性脂肪变。结论适当的高脂高胆固醇配比饮食是一种快速有效的脂肪肝模型制作方法 。
OBJECTIVE: To investigate the optimal diet formulation for animal model of non-alcoholic fatty liver in mice and provide valuable experimental data for further research on fatty liver and its treatment. Methods 80 KM mice were randomly divided into 4 groups, A, B, C model group, group D as the control group. The model group was given different gradient of high-fat diet, the control group was given a normal diet. Five rats in the model group were sacrificed every 1 week after the model was established to observe the formation of the pathological model, blood test and biopsy were performed. The biochemical indexes were detected by automatic biochemical analyzer. The liver, liver homogenate, Measured LTG, LTC. Results During modeling, the body weight of model group B was faster than that of normal control group and model group A and C groups. At the end of the experiment, the body weight of group A was 7.62% on average, the weight of group B was 16.76% on average, and the weight of group C was 11.14% on average. The body weight and liver index of model group were significantly higher than those of normal group (P <0.01). The biochemical indexes showed that the levels of TC, TG, ALT, AST, LDL-C, LTG and LTC in the model group decreased to a certain extent (P <0.01) , Among which the change of group B was the largest (P <0.01). In pathology, mice in group B developed mild hepatic steatosis as early as 1 week after modeling, and mice in group B exhibited moderate steatosis at 2 weeks. 4 weeks group B mice were moderate to severe bullous steatosis. Conclusion The appropriate high-fat and high-cholesterol diet is a fast and effective method for making fatty liver model.
