目的研究碱性成纤维细胞生长因子(b FGF)对染铅大鼠海马神经元细胞外调节蛋白激酶(ERK)的保护作用。方法神经元培养7 d后,用0.6μg/L的b FGF以及不同浓度铅处理细胞,然后测定ERK活力。结果铅处理神经元30 min后,在铅浓度0.1~2.0μmol/L时ERK活性明显升高,其中2.0μmol/L时最高。经过b FGF处理以后的细胞,在铅浓度1.0~4.0μmol/L时ERK活性与对照组相比无明显差异。结论染铅浓度不同对ERK活性影响也不同,b FGF可以保护神经元中ERK活性。
Objective To investigate the protective effect of basic fibroblast growth factor (b FGF) on extracellular regulated protein kinase (ERK) in hippocampal neurons of lead-exposed rats. Methods After cultured for 7 days, the cells were treated with 0.6 μg / L b FGF and different concentrations of Pb, and the activity of ERK was measured. Results After exposed to lead for 30 min, ERK activity was significantly increased at lead concentrations of 0.1-2.0 μmol / L and highest at 2.0 μmol / L. The cells treated with b FGF had no significant difference in the activity of ERK at the concentration of 1.0 ~ 4.0μmol / L compared with the control group. Conclusion Different concentrations of lead exposure have different effects on ERK activity. B FGF can protect the activity of ERK in neurons.
