目的通过小鼠眼球标本的制片质量和染色效果的比较,筛选最佳固定液。方法取健康成年小鼠眼球400只,随机分为5组,分别投入中性甲醛钙液、甲醛-乙醇固定液(Verhoeff液)、重铬酸钾-甲醛-冰醋酸-三氯醋酸-醋酸钠水溶液固定液(Kolmer液)、甲醛-冰醋酸-乙醇固定液(FAA液)、甲醛-冰醋酸-乙醇-生理盐水混合固定液(改良FAA液),按8 h、12 h、24 h及48 h固定,每个时间段取出20只,常规制片、苏木素-伊红(HE)染色,显微镜下观察。结果 5种固定液对小鼠眼球标本都有固定作用。Kolmer液固定效果最差:优级片0张(0%),中级片1张(5%),差级片19张(95%),不适用于眼球标本的固定。改良FAA液固定效果最好:优级片19张(95%),中级片1张(5%),差级片0张(0%),且以固定24 h最佳。结论改良FAA液固定小鼠眼球,标本的制片质量和染色效果最好,适用于临床和科研。
Objective To screen the best fixative by comparing the preparation quality and staining effect of mouse eyeball specimens. Methods 400 healthy adult mice were randomly divided into 5 groups: normal formaldehyde solution, formalin - ethanol solution (Verhoeff solution), potassium dichromate - formaldehyde - glacial acetic acid - trichloroacetic acid - sodium acetate (Kolmer solution), formaldehyde-glacial acetic acid-ethanol fixation solution (FAA solution), formaldehyde-glacial acetic acid-ethanol-saline mixed solution h fixed, each time out of 20, conventional production, hematoxylin-eosin (HE) staining, microscopic observation. Results The five kinds of fixatives had a fixed effect on mouse eyeball specimens. Kolmer has the worst liquid fixation effect: 0 (0%) for premium, 1 for middle (5%) and 19 (95%) for differential and is not suitable for the fixation of eye specimens. The modified FAA solution had the best effect of fixation: 19 excellent (95%), 1 intermediate (5%) and 0 differential (0%). Conclusion Improved FAA liquid-fixed mouse eyeballs, the preparation of the best quality and staining results for clinical and scientific research.
