以山药为原料,经分离提取纯化等一系列步骤得到山药水溶性多糖,用Sephadex G-100柱层析,发现经DEAE层析后的多糖为均一组分,其比旋光度为+162.5°,属于水溶中性多糖。经薄层层析以及GC-MS分析测定该中性糖为葡萄糖和甘露糖组成,其摩尔比为0.56:0.44。红外光谱和NMR谱分析显示该中性糖有α-异构体吡喃己糖环,它们归属为α-D-葡萄糖和α-D-甘露糖。
Taking yam as raw material, the yam water-soluble polysaccharide was obtained through a series of steps such as separation, extraction and purification, and the polysaccharide was homogenized by DEAE chromatography with Sephadex G-100 column chromatography. The specific rotation was + 162.5 °, Belong to water-soluble neutral polysaccharide. The neutral sugars were composed of glucose and mannitol as determined by thin layer chromatography and GC-MS analysis at a molar ratio of 0.56: 0.44. Infrared spectroscopy and NMR spectral analysis showed that the neutral sugars had the α-isomeric hexopyranose ring, which belonged to α-D-glucose and α-D-mannose.
