本研究建立一种快速、直接的甲胎蛋白(AFP) 变异体分析法,这种新方法“交叉亲和酶联免疫电泳分析法”是由小扁豆凝集素(LCA)亲和电泳和酶联免疫电泳结合构成的,能直接将AFP定量地分为LCA反应性(LCA-R)和非反应性(LCA-N) 两个部分。本法的优点是灵敏度高 (1~2ng AFP即可分析),测定快速(6~7小时),图形十分清晰,试剂稳定期很长。47例病理确诊的原发性肝癌患者血清AFP LCA-R≥25%者占91.5%;46例良性肝病为2.2%,具有良好的鉴别诊断价值。
This study establishes a rapid and direct analysis of alpha-fetoprotein (AFP) variants. This new method, “Cross-Affinity Enzyme-linked Immunoelectrophoretic Assay,” is performed by lentil agglutinin (LCA) affinity electrophoresis and enzyme-linked immunoassay. The combination of immunoelectrophoresis and composition can directly divide AFP into two parts: LCA-reactive (LCA-R) and non-reactive (LCA-N). The advantages of this method are high sensitivity (1 to 2 ng AFP can be analyzed), rapid measurement (6 to 7 hours), very clear graphics, and long reagent stability. Forty-seven patients with pathologically confirmed primary liver cancer had serum AFP LCA-R≥25% accounting for 91.5%; 46 patients with benign liver disease had 2.2%, which had good differential diagnosis value.
