目的 观察 8 Br cAMP对HL 6 0细胞生长、分化及其相关基因的调节。方法 组织化学、原位杂交和完整细胞RNA斑点印迹。结果 8 Br cAMP作用 2 4h ,HL 6 0细胞c fos癌基因和p2 1waf1抑癌基因表达明显升高 ,c myc癌基因表达明显降低 ;8 Br cAMP作用 48h ,HL 6 0细胞生长、增殖明显受抑制 ,并向中性粒细胞方向分化。结论 8 Br cAMP可调整HL 6 0细胞生长、分化及相关癌基因和抑癌基因的表达 ,抑制HL 6 0细胞的生长 ,促进HL 6 0细胞的分化
Objective To observe the effect of 8 Br cAMP on the growth, differentiation and related genes of HL 60 cells. Methods Histochemistry, in situ hybridization and whole cell RNA dot blot. Results 8 Br cAMP for 24 h, the expression of c fos oncogene and p2 1waf1 tumor suppressor gene in HL 60 cells was significantly increased, and the expression of c myc oncogene was significantly decreased. 8 Br cAMP for 48 h, the growth and proliferation of HL 60 cells were significantly affected. Inhibit and differentiate towards neutrophils. Conclusion 8 Br cAMP can regulate the growth and differentiation of HL 60 cells and the expression of related oncogenes and tumor suppressor genes, inhibit the growth of HL 60 cells, and promote the differentiation of HL 60 cells.
