环磷酰胺(cyclophosphamide,Cy)具有致畸作用。本文采用紫外吸收法测定大鼠胚胎DNA含量,观察Cy在给药后不同时间对DNA合成的抑制作用,并通过组织染色技术观察Cy对胚胎神经上皮细胞有丝分裂的影响。 未经产Sprague-Dawley?大鼠,体重240±SD20g,随机分为5组,于妊娠d13一次ipCy15mg/kg,分别于给药后4,8,12,24,48h处死取材。将胚胎制成匀浆,用氯化钠及氯仿-异戊醇提取DNA,751-G型
Cyclophosphamide (Cy) has a teratogenic effect. In this paper, the DNA content of rat embryos was determined by ultraviolet absorption method. The inhibitory effect of Cy on DNA synthesis was observed at different times after administration. The effect of Cy on the mitosis of embryonic neural epithelial cells was observed by tissue staining. Sprague-Dawley rats without body weight 240 ± SD20g, were randomly divided into 5 groups, once in pregnancy d13 ipCy15mg / kg, were sacrificed at 4,8,12,24,48h after administration. The embryos were homogenized, DNA was extracted with sodium chloride and chloroform-isoamyl alcohol, type 751-G
