目的:研究袪脂保肝颗粒对高脂饮食所致脂肪肝大鼠的防治效果及机制。方法:采用高脂饲料连续12周复制大鼠脂肪肝模型,期间分别对不同组别大鼠进行相应药物干预,观察袪脂保肝颗粒(24,12,6 g.kg-1)连续ig 12周对脂肪肝的防治作用,并初步探讨其疗效机制。结果:与空白对照组相比,模型组肝组织胆固醇(CHO),甘油三酯(TG)水平有显著升高(P<0.01),血清天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)显著升高,肝脏病理呈脂肪性肝病变,并伴有明显细胞损伤和组织坏死;与模型组比较,祛脂保肝颗粒各剂量组肝TG,CHO均显著降低(P<0.05或P<0.01);血清AST,ALT显著降低,肝脏病理损伤减轻。结论:袪脂保肝颗粒对脂肪肝具有较好的防治作用,可通过抗氧化机制减轻肝细胞损伤。
Objective: To study the preventive and therapeutic effects and mechanism of Daozhi Baogan Granules on fatty liver rats induced by high-fat diet. Methods: The model of fatty liver was duplicated in high-fat diet for 12 weeks. Corresponding drug intervention was given to rats in different groups. The effects of Zhi Zhi Baogan Granule (24, 12, 6 g.kg-1) Week on the prevention and treatment of fatty liver and preliminary study of its mechanism of action. Results: Compared with the blank control group, the levels of cholesterol (CHO) and triglyceride (TG) in the model group were significantly increased (P <0.01), the serum aspartate aminotransferase (AST), alanine ALT was significantly increased, and hepatic pathology was fatty liver disease accompanied by obvious cell injury and tissue necrosis. Compared with model group, the hepatic TG and CHO of each group were significantly decreased (P <0.05 or P <0.01). Serum AST, ALT were significantly decreased, and liver pathological damage was alleviated. CONCLUSION: SMG can effectively prevent and cure fatty liver and reduce the damage of liver cells through anti-oxidative mechanism.
