目的探讨三氧化二砷(As2O3)在低氧条件下对体外培养的人胃癌细胞SGC-7901生长的抑制及凋亡影响。方法用氯化钴(CoC l2)制作低氧模型,设常氧组、低氧组、低氧加药物组,分别取0.5、1.0、2.0、5.0、10.0μmol/L五个不同浓度的As2O3作用于胃癌细胞,用细胞活力测定(MTT)法检测药物作用后的细胞活力,HOECHST33258染色试剂盒测细胞凋亡。结果①低氧加药物组As2O3对SGC-7901细胞生长有抑制作用,与常氧对照组比较差异有统计学意义(P<0.05),并呈剂量-时间-效应关系。②低氧加药物组中,随着As2O3浓度的升高,胃癌细胞凋亡率也逐渐升高(P<0.05)。结论低氧下As2O3对人胃癌细胞SGC-7901生长有抑制和诱导凋亡作用,这种作用可能是As2O3发挥抗肿瘤的生物学基础。
Objective To investigate the effects of arsenic trioxide (As2O3) on the growth and apoptosis of human gastric cancer cell line SGC-7901 under hypoxic conditions. Methods Hypoxic model was made with cobalt chloride (CoCl 2). The normoxia group, hypoxia group and hypoxia plus drug group were respectively treated with 0.5, 1.0, 2.0, 5.0 and 10.0 μmol / L As2O3 In gastric cancer cells, cell viability was detected by MTT assay and apoptosis was detected by HOECHST33258 staining kit. Results ① Hypoxia and As2O3 inhibited the growth of SGC-7901 cells, and the difference was statistically significant (P <0.05) compared with normoxia control group, and showed a dose-time-effect relationship. ② In hypoxia plus drug group, the apoptosis rate of gastric cancer cells also increased gradually with the increase of As2O3 concentration (P <0.05). Conclusion As2O3 can inhibit the growth of human gastric cancer cell SGC-7901 and induce apoptosis in hypoxia. This effect may be the biological basis of anti-tumor effect of As2O3.
