从鲨鱼肝中分离纯化了鲨鱼肝刺激物质(sHSS),并对其理化性质进行了研究。纯化过程包括匀浆、离心、超滤、DEAE-sephadex A25 层析、FPLC mono Q 层析。经此过程, sHSS 活性可提高到9000IU/mg,纯化倍数可达750倍。经SDS聚丙烯酰胺凝胶电泳和高效凝胶过滤检测分别只出现一条带和一个洗脱峰。用质谱法测其分子量为16973Da。用PE-ABD491A蛋白质N-末端测序仪测定其N-末端氨基酸排列顺序是N-Met-Arg-Thr-Gln-Glu-His-Thr- Lys-Ser-Val。sHSS具有热稳定性和耐酸碱性。
Shark liver stimulant (sHSS) was isolated and purified from shark liver and its physicochemical properties were studied. The purification process included homogenization, centrifugation, ultrafiltration, DEAE-sephadex A25 chromatography, and FPLC mono Q chromatography. After this process, the sHSS activity can be increased to 9000 IU/mg, and the purification factor can reach 750 times. Only one band and one elution peak were detected by SDS polyacrylamide gel electrophoresis and high-performance gel filtration. Its molecular weight measured by mass spectrometry was 16973 Da. The N-terminal amino acid sequence of the PE-ABD491A protein N-terminal sequencer was N-Met-Arg-Thr-Gln-Glu-His-Thr-Lys-Ser-Val. sHSS has thermal stability and acid and alkali resistance.
