目的:建立高效液相色谱法测定小槐花药材中山柰酚的含量的方法。方法:样品用甲醇-25%盐酸(4∶1)混合溶液回流提取1.5 h,用Thermo BDS Hyperl C18柱(4.6 mm×250 mm,5μm)色谱柱分离,以乙腈-0.2%磷酸溶液(30∶70)为流动相,流速1.0 mL.min-1,柱温30℃,检测波长368 nm,进样量10μL,以外标法定量。结果:山奈酚进样量在18.94~189.4 ng与峰面积值呈现良好的线性关系(r=0.999 6),平均回收率为105.5%,RSD 1.2%(n=6)。结论:该法操作简便,结果准确可靠,重复性及稳定性良好,可作为控制小槐花药材质量的方法。
Objective: To establish a method for the determination of kaempferol in Xiaohuai medicinal materials by high performance liquid chromatography. Methods: The sample was extracted with a mixed solution of methanol and 25% hydrochloric acid (4: 1) for 1.5 h and separated on a Thermo BDS Hyperl C18 column (4.6 mm × 250 mm, 5 μm) with acetonitrile-0.2% phosphoric acid 70) as the mobile phase at a flow rate of 1.0 mL · min-1. The column temperature was 30 ℃, the detection wavelength was 368 nm, and the injection volume was 10 μL. Results: The linear range of kaempferol injection was 18.94 ~ 189.4 ng (r = 0.999 6). The average recoveries were 105.5% and RSD 1.2% (n = 6). Conclusion: The method is simple, accurate and reliable, reproducible and stable, which can be used as a method to control the quality of Xiaohuaihua medicinal materials.
