了解L7212白血病小鼠及其受体鼠——近交系615小鼠肿瘤坏死因子α(TNF-α)活性及TNF-αmRNA的表达,以及清热解毒中药清毒饮对TNF-α活性及TNF-αmRNA表达的影响。用WEHI-164细胞株及细胞原位杂交的方法检测了由伴刀豆凝集素A(Con A)诱导的脾细胞培养上清中TNF-α活性及脾细胞TNF-αmRNA表达。由ConA诱导的L7212白血病小鼠脾细胞培养上清中TNF-α活性明显低于615小鼠,P<0.01;其脾细胞中TNF-α mRNA表达也明显低于615小鼠,P<0.01。清毒饮能显著提高Con A诱导的L7212白血病小鼠脾细胞培养上清中的TNF-α活性,P<0.001,并能显著提高其mRNA表达水平,P<0.001。实验结果提示清毒饮可作为生物反应调节剂用于白血病的临床治疗。
To investigate the activity of tumor necrosis factor-α (TNF-α) and the expression of TNF-α mRNA in L7212 leukemia mice and their recipient mice--inbred strain 615 mice, and to investigate the effects of Qingrejiedu traditional Chinese medicine Qingduyin on TNF-α activity and TNF-α Effect of α mRNA expression. The activity of TNF-α and the expression of TNF-α mRNA in splenocytes of culture supernatants of splenocytes induced by concanavalin A (Con A) were detected by WEHI-164 cell line and in situ hybridization. The activity of TNF-α in the culture supernatant of spleen cells of L7212 leukemia mice induced by ConA was significantly lower than that of 615 mice (P<0.01), and the expression of TNF-α mRNA in spleen cells was also significantly lower than that of 615 mice (P<0.01). Qing Du Yin can significantly increase the activity of TNF-α in the supernatant of Con A-induced splenocytes of L7212 leukemia mice, P<0.001, and can significantly increase its mRNA expression level, P<0.001. The experimental results suggest that Qingduyin can be used as a biological response modifier for the clinical treatment of leukemia.
