目的观察活络健骨胶囊的实验镇痛效应。方法分别采用扭体、电刺激及热板法三种镇痛试验。每种实验又将60只小鼠随机分成6组并给予不同药物灌胃:A组(空白对照),生理盐水0.25mL/10g;B组(阳性对照),双氯芬酸3.5mg/kg;C组(阳性对照),哌替啶15mg/kg;D、E、F组分别为试药低、中、高剂量活络健骨胶囊(剂量分别为14、21、28g/kg)。然后在预处理后的不同时间分别观察药物对化学、电极热板刺激痛反应的影响。结果活络健骨胶囊低、中、高剂量组均能抑制化学刺激鼠扭体反应,与空白组比较,低剂量组显效(P<0.01);中、高剂量组作用更强(P<0.001)。对于电刺激鼠嘶叫反应,与空白组比较,低剂量组有效(P<0.05);中、高剂量组显效(P<0.01)。亦能明显延长小鼠对热板刺激引起痛反应的时间,各试药组与空白对照组比较(P<0.05);活络健骨胶囊与B、C组之间在30、60、120min镇痛作用基本相同(P>0.05),但在180min时阳性对照药不及活络健骨胶囊的效应(P<0.05)。结论活络健骨胶囊具有良好镇痛作用,其作用比哌替啶(小鼠灌胃)慢而持久,其镇痛强度大于双氯芬酸。
Objective To observe the experimental analgesic effect of Huoluo Jiangu capsule. Methods Three types of analgesic tests were used: writhing, electrical stimulation and hot plate method. In each experiment, 60 mice were randomly divided into 6 groups and given different drugs by gavage: group A (blank control), normal saline 0.25 mL/10 g; group B (positive control), diclofenac 3.5 mg/kg; group C ( Positive control), pethidine 15mg/kg; D, E, F group were low-, medium- and high-dose Huoxue Jiangu capsules (14, 21, and 28 g/kg, respectively). Then at different time after pretreatment, the effect of the drug on chemical and electrode hot plate stimulated pain was observed. Results Huoluo Jiangu capsule low, middle and high dose group can inhibit the writhing response of chemical stimulation rats, compared with the blank group, the low dose group markedly (P<0.01); the high dose group has a stronger effect (P<0.001) . For electrical stimulation of rat squeak response, compared with the blank group, the low dose group was effective (P<0.05); the middle and high dose groups were markedly effective (P<0.01). It also significantly prolonged the time of mouse pain induced by hot plate stimulation. Each drug group was compared with the blank control group (P<0.05). Analgesic effect was observed at 30, 60, and 120 minutes between Huoluo Jiangu capsule and B and C groups. The effects were basically the same (P>0.05), but the positive control drug was less than that of Huoluo Jiangu Capsule at 180 minutes (P<0.05). Conclusion Huoluo Jiangu capsule has good analgesic effect, and its effect is slower and longer than pethidine (mice gavage). Its analgesic intensity is stronger than that of diclofenac.
