OBJECTIVE To introduce a novel surrogate analyte approach for definitive quantitation of endogenous drugs using LC-MS-based technology in biological samples.METHODS We searched PubMed for articles published with general search terms of ″LC-MS″ and ″surrogate analyte″,the literatures were further investigated,analyzed and summarized.RESULTS Detailed discussion will be given for the basic concept of the new approach,development and optimization steps of the analytical procedure and important considerations that should be pay attention during every steps,and make a few suggestions.CONCLUSION Because of the advantages of sensitive,accurate,simple and high-throughput compared with conventional methods,such as standard addition and surrogate matrix methods,surrogate analyte strategy using LC-MS based approach will undoubtedly become the preferred method for definitive quantitation of endogenous drugs in the future.
