使用聚丙烯酰胺凝胶垂直平板电泳法 ,研究了不同浓度 (10 - 4~ 10 - 8mol/L)的Cd2 + ,Ni2 +和Hg2 + 对大麦根尖细胞过氧化物酶同工酶的影响。结果表明 ,10 - 4mol/LCd2 + 和Hg2 + 处理 2 4h均比对照增加 1条酶带 (Rf=0 .36 ) ,但前者缺失了 1条Rf 值为 0 80的酶带 8。处理 4 8h后 ,10 - 6mol/L的Hg2 + 处理组又增加 1条Rf 值为 0 36的新酶带。其余浓度的Cd2 + ,Hg2 + 和 3种浓度的Ni2 + 均可使同工酶强弱发生改变。Cd2 + 和Hg2 + 对过氧化酶的影响明显大于Ni2 + 。 3种重金属对大麦过氧化物酶同工酶的影响与浓度和处理时间有关
The effects of Cd2 +, Ni2 + and Hg2 + with different concentrations (10-4 ~ 10-8 mol / L) on the peroxidase isoenzyme of barley root tip cells were studied by polyacrylamide gel electrophoresis. The results showed that one band (Rf = 0.36) increased by 10 -4 mol / L Cd2 + and Hg2 + for 24 h compared with the control, but the former lacked a band of 8 with Rf value of 0 80. After 48 h of treatment, a new enzyme band with an Rf value of 0 36 was added to the 10 - 6 mol / L Hg2 + treatment group. The remaining concentrations of Cd2 +, Hg2 + and three concentrations of Ni2 + can make the isozyme strength changes. The effect of Cd2 + and Hg2 + on peroxidase was significantly greater than that of Ni2 +.
