目的通过检测结肠癌组织及其癌旁组织中磷酸肌醇特异性的磷脂酶Cε1(PI-PLCε1)的表达情况,探讨PI-PLCε1在结肠癌发病过程中的临床意义。方法采用实时荧光定量PCR(real-time quantitative PCR,qRT-PCR)及免疫组化的方法检测42例结肠癌组织及其相对应的癌旁组织中PI-PLCε1的表达情况,并分析肿瘤分化程度及肿瘤部位对于结肠癌组织中PI-PLCε1表达情况的影响。结果 qRT-PCR结果显示,结肠癌组织中PI-PLCε1基因的表达低于其癌旁组织(P<0.05),不同肿瘤分化程度及肿瘤部位结肠癌组织中PIPLCε1基因的表达差异没有统计学意义(P>0.05)。免疫组化结果显示,结肠癌组织中PI-PLCε1蛋白的阳性表达率低于其癌旁组织(P<0.05)。不同肿瘤分化程度结肠癌组织中PI-PLCε1蛋白的阳性表达率差异没有统计学意义(P>0.05),但不同肿瘤部位中PI-PLCε1蛋白的阳性表达率差异有统计学意义(P<0.05)。结论 PI-PLCε1在结肠癌组织中的表达显著减少,且与肿瘤分化程度无关。
Objective To investigate the clinical significance of PI-PLCε1 in the pathogenesis of colon cancer by detecting phosphosoinosine-specific phospholipase Cε1 (PI-PLCε1) expression in colon cancer tissues and adjacent tissues. Methods Real-time quantitative PCR (qRT-PCR) and immunohistochemistry were used to detect the expression of PI-PLCε1 in 42 cases of colorectal cancer tissues and corresponding paracancerous tissues, and the degree of tumor differentiation And tumor site on the expression of PI-PLCε1 in colon cancer tissue. Results The results of qRT-PCR showed that the expression of PI-PLCε1 in colon cancer tissues was lower than that in adjacent tissues (P <0.05). There was no statistical difference in the differentiation degree of tumor tissues and the expression of PILCLCε1 in colon cancer tissues Significance (P> 0.05). The results of immunohistochemistry showed that the positive expression rate of PI-PLCε1 protein in colon cancer tissues was lower than that in adjacent tissues (P <0.05). There was no significant difference in the positive expression rate of PI-PLCε1 protein in colon cancer with different tumor differentiation (P> 0.05), but there was significant difference in the positive expression rates of PI-PLCε1 between different tumor sites (P <0.05) . Conclusion The expression of PI-PLCε1 in colon cancer tissues is significantly reduced, and has no relation with the degree of tumor differentiation.
