目的:探讨RASSF1A(RAS相关区域家族1A)基因转录表达与子宫内膜癌发生、发展的关系。方法:采用逆转录-聚合酶链反应(PT-PCR)方法检测50例子宫内膜癌组织及20例正常子宫内膜组织中RASSF1A mRNA的表达水平。结果:①RASSF1A mRNA在所有正常子宫内膜组织中表达,而在子宫内膜癌组织中存在较高的表达缺失率(54%);②RASSF1A mRNA的表达缺失与子宫内膜癌手术病理分期、淋巴结转移、肌层浸润及病理组织学类型有明显相关性(P<0.05);RASSF1A mRNA表达缺失与年龄和病理分级未见明显相关(P>0.05)。结论:RASSF1A转录表达缺失可能参与调控子宫内膜癌的发生、发展过程,并进而影响其生物学特点及预后。
Objective: To investigate the relationship between the transcriptional expression of RASSF1A (RAS-related region family 1A) gene and the occurrence and development of endometrial carcinoma. Methods: The expression of RASSF1A mRNA in 50 cases of endometrial carcinoma and 20 cases of normal endometrium was detected by reverse transcription-polymerase chain reaction (PT-PCR). RESULTS: (1) RASSF1A mRNA was expressed in all normal endometrial tissues, but not in endometrial carcinoma (54%). ② The loss of RASSF1A mRNA expression correlated with the pathological stage, lymph node metastasis (P <0.05). There was no significant correlation between the expression of RASSF1A mRNA and age and pathological grade (P> 0.05). Conclusion: The lack of transcriptional expression of RASSF1A may be involved in the regulation of endometrial cancer, the development of endometrial cancer and its biological characteristics and prognosis.
