目的:比较整合素α6mRNA在原发性肝细胞肝癌(HCC)癌组织及正常肝组织中表达的差异,分析整合素α6mRNA的表达与HCC的侵袭程度、远处转移、临床分期之间的关系。方法:取肝癌患者手术切除标本,RT-PCR扩增整合素α6mRNA后对扩增产物进行电泳及图象分析。结果:癌组织、癌旁组织与相应正常肝组织中整合素α6mRNA定性表达阳性率差异有统计学意义(P<0.001)。但癌旁组织和正常肝组织之间整合素α6mRNA表达阳性率差异无统计学意义(P=0.4862)。在表达阳性的癌组织中,其半定量分析显示无包膜者高于有包膜者,有肝内或肝外转移者高于无肝内或肝外转移者。随着Edmondson分级增高,整合素α6及β1mRNA表达量不断增高(P<0.001)。结论:整合素α6mRNA在HCC癌组织、癌旁组织和相应正常肝组织中均有阳性表达。但在HCC癌组织中表达明显高于其他组织。HCC癌组织中整合素α6mRNA表达高低与侵袭程度、有无肝内或肝外转移及临床分期密切相关。高表达的整合素α6mRNA预示着肝癌的高度恶性、高侵袭性、高转移性和预后不良。整合素α6mRNA可以作为反映HCC转移和侵袭特性的分子标记物。
OBJECTIVE: To compare the expression of integrin α6 mRNA in primary hepatocellular carcinoma (HCC) and normal liver tissues, and to analyze the relationship between the expression of integrin α6 mRNA and the invasive degree, distant metastasis and clinical stage of HCC. Methods: The specimens of patients with hepatocellular carcinoma were removed surgically. The products of integrin α6 mRNA were amplified by RT-PCR and analyzed by electrophoresis and image analysis. Results: The positive rates of integrin α6 mRNA expression in cancer tissues, adjacent non-cancerous tissues and corresponding normal liver tissues were significantly different (P <0.001). However, there was no significant difference in the positive rate of integrin α6 mRNA between adjacent non-cancerous tissues and normal liver tissues (P = 0.4862). In the positive expression of cancer tissue, semi-quantitative analysis showed that those without capsule were higher than those with capsule, with intrahepatic or extrahepatic metastasis than those without intrahepatic or extrahepatic metastasis. As Edmondson grade increased, the expression of integrin α6 and β1 mRNA increased (P <0.001). Conclusion: Integrin α6 mRNA is positively expressed in HCC, paracancerous tissues and corresponding normal liver tissues. However, the expression in HCC tissues was significantly higher than that in other tissues. The expression of integrin α6 mRNA in HCC tissues is closely related to the degree of invasiveness, intrahepatic or extrahepatic metastasis and clinical stage. Highly expressed integrin α6 mRNA predicts highly malignant, highly invasive, metastatic and poor prognosis of HCC. Integrin α6 mRNA can be used as a molecular marker reflecting the metastatic and invasive properties of HCC.
