以发根农杆菌A4、C58C1和A1476为诱导菌株,探讨外植体类型、发根农杆菌种类、侵染时间、预培养时间、菌液浓度和共培养天数对龙葵毛状根诱导率的影响。结果表明:外植体以叶片诱导率最高;发根农杆菌A4、C58C1和A1476侵染龙葵叶片均能诱导出毛状根,C58C1诱导率较高;较佳侵染时间为5min;发根农杆菌浓度在OD600=0.6时诱导效果较好;预培养和共培养时间均为2d时诱导率较高。该试验为大量生产毛状根,并利用毛状根培养技术生产药物龙葵碱提供了试验基础。
Agrobacterium rhizogenes A4, C58C1 and A1476 were used as induced strains. The effects of explant type, Agrobacterium rhizogenes, infection time, pre-culture time, bacterial concentration and co-cultivation days on the induction rate of hairy root of Solanum nigrum influences. The results showed that the explants had the highest rate of leaf induction; the hairy roots were induced by Agrobacterium rhizogenes A4, C58C1 and A1476, and the induction rate of C58C1 was higher. The optimal infection time was 5 min. Agrobacterium concentration of OD600 = 0.6 when the induction effect is better; pre-culture and co-culture time are 2d higher induction rate. This test provides the experimental basis for the mass production of hairy roots and the use of hairy root culture techniques for the production of solanine.
