目的 克隆肾癌相关新基因GYLZ RCC18的全长 ,并探讨其功能。方法 应用SMARTRACE技术克隆GYLZ RCC18的全长 ,应用逆转录PCR的方法检测其在肾癌组织和细胞系中的表达特点 ,以及与肾癌组织分级、分期的关系 ,通过转染GYLZ RCC18的反义寡核苷酸阻断其表达 ,观察其对肾癌细胞系生命特征的变化。结果 GYLZ RCC18全长约 3 5kb(基因号 :BE82 5 133)。GYLZ RCC18在肾癌特异高表达 ,正常肾表达量极低甚至不表达 ,两者表达量差别 1 9倍之间 ;GYLZ RCC18在高分级、高分期肾癌中表达明显高于低分级、低分期肾癌的表达。转染反义基因可明显抑制GRC 1的生长、降低其增殖活性、减少核分裂、并诱导其持续性凋亡。结论 本研究表明GYLZ RCC18是肾癌发生发展密切相关的重要新的癌基因 ,它的表达与肾癌细胞的生存、增殖、抗凋亡发展密切相关 ,它的成功克隆为今后肾癌分子机制研究提供了新的理论依据。
Objective To clone the full length of kidney cancer related gene GYLZ RCC18 and to explore its function. Methods The full length of GYLZ RCC18 was cloned by SMARTRACE technique. The expression of GYLZ RCC18 was detected by RT-PCR. The expression of GYLZ RCC18 in renal cell carcinoma was detected by SMARTRACE. Oligonucleotides block their expression and observe their changes in the life characteristics of renal cancer cell lines. Results GYLZ RCC18 was about 35kb in length (Gene No .: BE82 5 133). GYLZ RCC18 was highly expressed in renal cell carcinoma, and the expression of GYLZ RCC18 was significantly higher than that in low-grade, low-grade renal cell carcinoma Kidney cancer expression. Transfection of antisense gene can significantly inhibit the growth of GRC 1, reduce its proliferation activity, reduce mitosis, and induce its persistent apoptosis. Conclusions This study shows that GYLZ RCC18 is an important new oncogene closely associated with the development of renal cell carcinoma. Its expression is closely related to the survival, proliferation and anti-apoptosis of renal cell carcinoma. The successful cloning of GYLZ RCC18 is the molecular mechanism of future renal cell carcinoma Provided a new theoretical basis.
