目的 了解术后胃癌的多药耐药性与胃癌组织中P gp过量表达的关系。 方法 利用免疫组化方法 ,以 4种抗不同亚型P gp(C494,MRK16 ,JSB 1和C2 19)的单克隆抗体 (mAb) ,以及 4种不同抗P gpmAb的混合抗体和抗p5 3蛋白mAb ,分别检测 6 6例胃癌术后石蜡包埋组织中P gp和p5 3的表达 (其中 16例为冰冻切片 )。 结果 P gp呈中等或高度表达 (占 40 %~ 70 % )。依次为 :C494( 75 75 % ,5 0 /6 6 ) ,JSB 1( 6 2 12 % ,41/ 6 6 ) ,MRK16 ( 48 49% ,32 / 6 6 )和C2 19( 36 36 % ,2 4/ 6 6 )。 4种抗P gpmAb的混合物的阳性检出率为 78 79% ( 5 2 / 6 6 ) ,抗p5 3蛋白mAb的阳性检出率为6 0 6 1% ( 40 / 6 6 )。 4种抗P gpmAb的混合物与抗P gpmAbC494(G =0 90 ,P <0 0 0 0 0 1) ;抗P gpmAbC494与抗JSB 1mAb(G =0 6 0 ,P <0 0 0 0 0 5 ) ;抗MRK16mAb与抗C2 19mAb(G =0 6 4,P <0 0 0 0 1)均高度相关。石蜡和冰冻切片中P gpC494和JSB 1呈中度表达以上者阳性率基本一致 ;而C2 19和MRK16的阳性率相差则明显 (P <0 0 5 )。p5 3蛋白的表达与C494和JSB 1的表达相关性显著 (P <0 0 0 2和P <0 0 0 2 ) ,与MRK16和C2 19的表达也有相关性(P <0 0 1和P <0 0 5 )。结论 C494阳性表达率最高 ,可作为胃癌组织中耐药基?
Objective To investigate the relationship between multidrug resistance in gastric cancer and overexpression of P gp in gastric cancer. Methods Immunohistochemical staining was performed using four monoclonal antibodies (mAbs) against different subtypes of P gp (C494, MRK16, JSB 1 and C2 19) and four different anti-P gpmAb antibodies and anti-p5 3 mAb were used to detect the expression of P gp and p5 3 in paraffin-embedded tissues of 6 6 patients (16 of them were frozen sections). Results P gp was moderately or highly expressed (40% -70%). Followed by C494 (75 75%, 50/6 6), JSB 1 (62 12%, 41/6 6), MRK 16 (48 49%, 32/6 6) and C2 19 (36 36%, 2 4/6 6). The positive detection rate was 78 79% (5 2/6 6) for the 4 anti-P gpmAb mixtures and 6 0 6 1% (40/6 6) for the anti-p5 3 mAb. PgpmAbC494 (G = 090, P <0 0 0 0 0 1); anti-PgpmAbC494 and anti-JSB 1 mAb (G = 0 60, P 0 0 0 0 0 5) ; Anti-MRK16 mAb was highly correlated with anti-C219 mAb (G = 0 64, P <0 0 0 0 1). The positive rates of PgpC494 and JSB1 were moderately consistent in paraffin and frozen sections, while the positive rates of C219 and MRK16 were significantly different (P <0.05). The expression of p5-3 protein was significantly correlated with the expression of C494 and JSB1 (P <0.01 and P <0.002), and also with the expression of MRK16 and C219 (P <0.01 and P < 0 0 5). Conclusions C494 positive expression rate is the highest, which can be used as drug-resistant base in gastric cancer tissue.
