目的建立HPLC法同时测定莲子心提取物中莲心碱、异莲心碱和甲基莲心碱的含量。方法采用Hedera ODS-2色谱柱(4.6 mm×250 mm,5μm);流动相:甲醇-0.05%三乙胺溶液(72.5∶27.5);检测波长:283 nm;柱温:25℃;流速:1.0ml·min-1。结果 3种生物碱分离良好,莲心碱、异莲心碱、甲基莲心碱分别在3.92~117.60、3.84~115.20、5.44~163.20μg·ml-1范围内呈现良好的线性关系。利用该法测定莲子心提取物及其胶囊剂中生物碱的含量,平均总碱含量高于76%。结论该测定方法准确性高、重复性好、专属性高,可用于同时测定莲子心提取物及其胶囊剂中莲心碱、异莲心碱、甲基莲心碱的含量。
OBJECTIVE To establish a HPLC method for the simultaneous determination of lotus root, isolindine and neferine in the lotus root extract. Methods The mobile phase was methanol-0.05% triethylamine (72.5: 27.5). The detection wavelength was 283 nm. The column temperature was 25 ℃. The flow rate was 1.0 ml · min-1. Results The three alkaloids were well separated. The linear relationships of Liensinine, Isoproterenol and Neferine were linear in the range of 3.92-117.60, 3.84-115.20 and 5.44-1663.0μg · ml-1, respectively. The method was used to determine the content of alkaloids in the lotus root extract and its capsule, with the average total alkali content higher than 76%. Conclusion The method is accurate, reproducible and specific. It can be used for the simultaneous determination of lotus root, isolindine and neferine in lotus seed extract and its capsule.
