目的通过对复方龙脉宁各有效组分组成进行不同配比,探讨复方龙脉宁对寒凝血瘀大鼠体外血栓形成的影响。方法采用皮下注射肾上腺素加冰水浴法建立寒凝血瘀大鼠模型,用正交试验设计筛选各有效组分的配比组,对大鼠颈总动脉取血,用体外血栓仪测血栓长度、血栓干重。结果优选组合为A3B1C1D3能明显抑制寒凝血瘀大鼠体外血栓形成。结论正交试验设计与药效学实验相结合,可作为有效的研究复方药最佳组合与配比的方法。
OBJECTIVE To study the effect of compound Longmaining on thrombosis in cold coagulation and blood stasis rats in vitro by different proportions of the effective components of compound Longmaining. Methods Hypoxic-ischemic-stasis rat models were established by subcutaneous injection of epinephrine and ice-water bath. Orthogonal design was used to screen the ratio of effective components. Blood samples were collected from the common carotid artery in rats. Thrombus length, Thrombus dry weight. Results The optimal combination of A3B1C1D3 could significantly inhibit thrombosis in vitro in cold-blooded rats. Conclusion The combination of orthogonal design and pharmacodynamics experiment can be used as an effective method to study the optimal combination and proportion of compound prescriptions.
